I've been through the manual but it doesn't seem to address this.

The drone obviously now has an upward sensor.... Is this utilised in any RTH mode (operator started, low battery or loss of signal) ?

The manual itself mentions the forward sensors and how it'll avoid objects (identical to the mavic 1) but makes no mention of the upward sensor.

In other words, if someone is flying through a tunnel and loses signal, will the drone actually detect the obstruction (and if so, will it stop, attempt to go round or something else?) or is the sensor inactive in that mode so it'll just ascend into the obstacle as with previous models?

I would hope that this sensor is always active, since only the side sensors are called out as being *only* active during Tripod or Active Track. Try it and report back :p
